ToggleLongevity has gone mainstream. A few years ago, talk of slowing down aging lived in research labs and the odd wellness podcast. Now it is in your feed, at the local IV lounge, and in conversations with friends who suddenly know what a mitochondrion is. Behind the buzzwords, though, there is some real and genuinely interesting science. Here is a grounded look at two of the names you keep hearing, without the hype.
The starting point is a simple idea. As we get older, some of the machinery inside our cells becomes less efficient. Two areas draw a lot of attention from researchers: the way cells produce and manage energy, and the molecules that help keep that process running smoothly. That is where the two trending terms come in.
NAD+ and cellular energy
The first is NAD+, short for nicotinamide adenine dinucleotide. It is a coenzyme found in every cell and plays central roles in energy metabolism and enzymes involved in DNA repair. It is not a peptide, even though wellness marketing often groups it with them. NAD metabolism changes with age, but the pattern is tissue-dependent and more complex than a universal decline in every part of the body. Researchers are therefore testing whether NAD-boosting strategies affect health, not merely whether they raise a laboratory measurement.
Human research so far mostly uses precursors, compounds the body converts into NAD+. In a 2018 randomized trial, nicotinamide riboside was well tolerated over the study period and increased NAD-related metabolites in healthy middle-aged and older adults (Martens and colleagues, 2018). The trial did not establish a general anti-ageing effect. Reviews describe strong preclinical evidence for NAD-boosting approaches while emphasizing that meaningful clinical benefits in humans remain uncertain.
MOTS-c and the mitochondrial angle
The second name is MOTS-c, a mitochondrial-derived peptide encoded in mitochondrial DNA and studied in energy metabolism. A widely cited 2021 Nature Communications study found that circulating MOTS-c changed with exercise in people and reported effects on muscle performance and age-related physical decline in mice (Reynolds and colleagues, 2021). The intervention evidence was therefore primarily preclinical. MOTS-c is experimental, is not an approved medicine, and the FDA has stated that it has not identified human exposure data for administered MOTS-c drug products.
So what should a sensible person take from all this? A few things. The science of ageing is real and moving quickly, and these are legitimate research directions rather than pure marketing. At the same time, the boldest promises are running ahead of the proof, especially in humans. Treating any of it as a guaranteed fountain of youth is a mistake.
There is also a slightly unglamorous truth worth repeating. The interventions with the strongest evidence for aging well are still the ordinary ones. Regular movement, decent sleep, a good diet, not smoking, and managing stress do more for your cellular health than any trending compound, and they are free. The exciting research on NAD+ and MOTS-c is best understood as a possible complement to those basics, not a shortcut around them.
It also pays to understand the different forms these trends take, because they are not equivalent. NAD+ is offered through IV services, injections and oral precursor supplements, but raising NAD-related biomarkers does not itself prove a longevity benefit. Oral precursors carry more published human research than experimental peptides such as MOTS-c. More intensive and more expensive does not automatically mean more effective, and unapproved research compounds should not be self-administered.
If you are curious enough to look further, ask what outcome was actually measured and whether the evidence came from people, animals or cells. Independent testing and accurate labeling can provide limited quality information, but neither establishes safety, effectiveness or suitability for personal use.
Ageing well is not about chasing every new trend. It is about understanding what the science actually says and keeping the fundamentals in place.
